Induced Electrostatic Charge in Poleward Motion of Chromosomes During Mitosis

چکیده

Previously, I have proposed a model for post-attachment chromosome motions based on electrostatics and assumptions regarding net charge on kinetochores and the pericentriolar material of centrosomes. The present work shows that given the electric dipole nature of tubulin, layered water associated with electric charge, and the dynamic instability of microtubules, it is possible to account for these motions within a comprehensive model based on electrostatics without making any assumptions regarding net charge on these structures – a minimal assumptions comprehensive model. Tubulin microtubule subunits are electric dipolar structures that can induce charge on a kinetochore or centrosome matrix through the reduced permittivity of layered cytoplasmic water. Thus, prometaphase

The force for poleward chromosome motion in Haemanthus cells acts along the length of the chromosome during metaphase but only at the kinetochore during anaphase

The force for poleward chromosome motion during mitosis is thought to act, in all higher organisms, exclusively through the kinetochore. We have used time-lapse. video-enhanced, differential interference contrast light microscopy to determine the behavior of kinetochore-free "acentric" chromosome fragments and "monocentric" chromosomes containing one kinetochore, created at various stages of mi...

Kinetochore-Independent Chromosome Poleward Movement during Anaphase of Meiosis II in Mouse Eggs

Kinetochores are considered to be the key structures that physically connect spindle microtubules to the chromosomes and play an important role in chromosome segregation during mitosis. Due to different mechanisms of spindle assembly between centrosome-containing mitotic cells and acentrosomal meiotic oocytes, it is unclear how a meiotic spindle generates the poleward forces to drive two rounds...

Mitosis in Tilia americana endosperm

The endosperm cells of the American basswood Tilia americana are favorable experimental material for investigating the birefringence of living plant spindles and anaphase movement of chromosomes. The behavior of the chromosomes in anaphase and the formation of the phragmoplast are unique. The numerous (3 n equals 123), small chromosomes move in precise, parallel rows until midanaphase when they...

Electrostatic force generation in chromosome motions during mitosis

In previous work, I have proposed that nanoscale electrostatics plays a significant role in aster (spindle) assembly and motion, and in force generation at kinetochores and chromosome arms for prometaphase, metaphase and anaphase-A motions during mitosis. I have also discussed the possible role of electrostatics in anaphase-B cell elongation. Recent experimental studies have revealed that force...

A Regulatory Switch Alters Chromosome Motions at the Metaphase to Anaphase Transition

To achieve chromosome segregation during mitosis, sister chromatids must undergo a dramatic change in their behavior to switch from balanced oscillations at the metaphase plate to directed poleward motion during anaphase. However, the factors that alter chromosome behavior at the metaphase-to-anaphase transition remain incompletely understood. Here, we perform time-lapse imaging to analyze anap...
